ABSTRACT

The intracranial compliance in type 2 diabetes mellitus (T2DM) patients and the association with cardiovascular autonomic control have not been fully elucidated. The aim of this study was to assess intracranial compliance using the noninvasive intracranial pressure (niICP) and the monitoring of waveform peaks (P1, P2, and P3) and the relationship with cardiovascular autonomic control in T2DM patients. Thirty-two men aged 40-60 years without cardiovascular autonomic neuropathy (CAN) were studied: T2DMG (n=16) and control group CG (n=16). The niICP was evaluated by a noninvasive extracranial sensor placed on the scalp. Cardiovascular autonomic control was evaluated by indices of the baroreflex sensitivity (BRS), from temporal series of R-R intervals of electrocardiogram and systolic arterial pressure, during supine and orthostatic positions. The participants remained in the supine position for 15 min and then 15 min more in orthostatism. T2DMG presented a decrease of the P2/P1 ratio during the orthostatic position (P<0.001). There was a negative moderate correlation between the P2 peak with cardiovascular coupling (K2HP-SAPLF) in supine (r=-0.612, P=0.011) and orthostatic (r=-0.568, P=0.020) positions in T2DMG. We concluded that T2DM patients without CAN and cardiovascular complications presented intracranial compliance similar to healthy subjects. Despite preserved intracranial adjustments, T2DM patients had a response of greater magnitude in orthostatism. In addition, the decoupling between the heart period and blood pressure signal oscillations in low frequency appeared to be related to the worsening of intracranial compliance due to the increased P2 peak.

ABSTRACT

Esta pesquisa avaliou a TIP e a dinâmica de anticorpos (ACs) específicos em bezerros naturalmente expostos aos agentes causadores da doença respiratória bovina (DRB). Foram selecionados 19 bezerros Holandeses alimentados com colostro proveniente de doadoras vacinadas para DRB. Amostras de soro foram obtidas antes e após a ingestão do colostro (48h) para a soroneutralização (SN). Os valores médios (log2) detectados após colostragem foram de 11,5±1,6 (BVDV), 8,8±1,3 (BoHV-1), 5,5±1,6 (BRSV) e 8,4±1,5 (BPIV-3). Cinco bezerros foram criados do nascimento aos 240 dias de vida, observando-se decréscimo nos títulos de ACs para BVDV, BoHV-1 e BPIV-3 ao longo do tempo (P≤0,001). As taxas de infecções detectadas entre o D14 e o D240 foram de 40% (2/5), 20% (1/5), 80% (4/5), e 60% (3/5), respectivamente, para BVDV, BoHV-1, BRSV e BPIV-3. A maioria dos bezerros manifestou broncopneumonia após as infecções virais. Os bezerros apresentaram ACs para todas as viroses às 48 horas de vida, porém os títulos adquiridos para o BRSV foram baixos. A susceptibilidade para as infecções variou de acordo com os níveis e a duração dos títulos de ACs maternos.(AU)


This research evaluated the PIT and the dynamics of specific antibody (Ab) for calves naturally exposed to the viral agents involved in Bovine Respiratory Disease (BRD). Nineteen Holstein calves fed colostrum from vaccinated donors for DRB. Serum samples were obtained before and after colostrum intake (48h) for serum neutralization (SN). Mean values (log2) detected after colostrum feeding were 11.5±1.6 (BVDV), 8.8 ±1.3 (BoHV-1) 5.5±1.6 (BRSV) and 8.4±1.5 (BPIV-3). Five calves were raised from birth to 240 days of life and presented a decrease in Ab titers for BVDV, BoHV-1 and BPIV-3 over time (P≤ 0.001). Infection rates from D14 to D240 were of 40% (2/5), 20% (1/5), 80% (4/5) and 60% (3/5), respectively for BVDV, BoHV-1, BRSV and BPIV-3. Most of the calves presented bronchopneumonia after seroconversion to the virus. Calves presented Ab for all viruses at 48 hours of life, however BRSV Ab titer were low. Levels and persistence of maternal antibody titers determined the susceptibility to viral infections.(AU)

ABSTRACT

The hematological and immunological development of calves from birth to 6 months of age was performed by hemogram and cellular phenotype. Ten male Holstein calves were assessed in 13 moments: before colostrum intake (D0), every 2 days until the 10th day of life (D2 to D10), at the 15th day after birth (D15), and then monthly up to 6 months (D180). Calves presented hemoconcentration on day (D) 0 and showed a gradual decrease in hematimetric rates until D180. The inversion of the neutrophilic for lymphocytic profile was observed on D4. The percentage of CD3+ cells on D10 was higher than D30 up to D180. The number of CD4+ and CD8+ cells did not change between time points. The number of CD21+ lymphocytes was significantly higher at early time points of D0 up to D15, compared on D30 until D150. In conclusion, the neonatal period was marked by stress leukogram in the first 4 days, and low number of B lymphocytes. These might be risk factors for bacterial infections responsible for navel inflammation and diarrhea. The increase in the number of B cells from 30 days of age demonstrated that the calves were functional and able to generate an immune response.(AU)


O desenvolvimento hematológico e o imunológico de bezerros do nascimento aos seis meses de idade foram avaliados por meio hemograma e imunofenotipagem dos linfócitos. Para tanto, 10 bezerros machos Holandeses foram avaliados em 13 momentos: antes da ingestão do colostro (D0), a cada dois dias até o 10º dia (D2 a D10), 15º dia (D15) e, em seguida, mensalmente até seis meses de idade (D180). Os bezerros apresentaram hemoconcentração em D0 e decréscimo gradual dos índices hematimétricos até D180. A inversão do perfil neutrofílico para linfocítico foi observada em D4. Os valores de CD3+ (%) em D10 foram maiores que aqueles observados em D30 até D180. CD4+ e CD8+ não apresentaram alterações entre os momentos. Os linfócitos B CD21+ aumentaram significativamente a partir dos primeiros momentos D0 até D15, em comparação com os momentos subsequentes D30 até D150. Em conclusão, o período neonatal foi caracterizado pelo leucograma de estresse nos primeiros quatro dias e o baixo número de linfócitos B. Esses fatos podem ser considerados como fatores de risco para infecções bacterianas responsáveis pelas inflamações umbilicais e pela diarreia. Por outro lado, o aumento de células B após os 30 dias de vida demonstrou que os bezerros foram capazes de gerar uma resposta imune.(AU)

O presente estudo foi conduzido para determinar o consumo e a digestibilidade dos nutrientes, bem como o comportamento ingestivo de cordeiros Somalis Brasileira, alimentados com diferentes níveis de energia metabolizável (EM). Foram utilizados 40 animais, não castrados, com peso vivo médio de 13,47±1,76kg. Os animais foram distribuídos em delineamento em blocos ao acaso, com cinco dietas com diferentes níveis de energia metabolizável (1,18; 2,07; 2,25; 2,42 e 2,69Mcal/kg de MS). O consumo de MS, PB, FDN, CF (g/dia), MS (g/kg0,75) e FDN (%PC e g/kg0,75) apresentaram efeito quadrático (P<0,05), assim como os coeficientes de digestibilidade (P<0,05) da PB, da FDN e dos CF. Foi averiguado efeito linear crescente (P<0,05) para digestibilidade da MS, da MO e dos CNF com o aumento da EM na dieta. Os tempos de alimentação e ócio (h/dia) foram influenciados pelos níveis de energia (P<0,05), e as eficiências de alimentação e ruminação (g FDN/h) apresentaram efeito quadrático (P<0,05). Conclui-se que o aumento dos níveis de EM das rações influencia o consumo e a digestibilidade dos nutrientes, bem como o comportamento ingestivo de cordeiros Somalis Brasileira...


The study was conducted to determine the intake, nutrient digestibility and ingestive behavior of Brazilian Somali lambs fed different levels of metabolizable energy (ME). Forty non-castrated Brazilian Somali lambs with average weight of 13.47±1.76kg were used. The animals were assigned to a randomized block design with eight replications per block and five diets with increasing metabolizable energy (1.18, 2.07, 2.25, 2.42 and 2.69 Mcal/kg DM). The quadratic effect (P<0.05) of the ME level was determined for DM, CP, NDF and FC (g/day) and DM (g/kg0,75) and NDF(%PC and g/kg0,75) intake, as well as the digestibility coefficients (P<0.05) for CP, NDF and CF. A linear effect (P<0.05) was observed for digestibility of DM, OM and NFC with the increase of ME in the diet. Eating and idle results (hours/day) were influenced by the energy levels (P<0.05) and efficiencies of feeding and rumination (g NDF/h) showed a quadratic effect (P<0.05). It is concluded that the increase of ME levels in the diet influences intatke, nutrient digestibility and ingestive behavior of Brazilian Somali lambs...

ABSTRACT

A preliminary study on xylitol production by Candida guilliermondii in sorghum straw hemicellulosic hydrolysate was performed. Hydrolysate had high xylose content and inhibitors concentrations did not exceed the commonly found values in other hemicellulosic hydrolysates. The highest xylitol yield (0.44 g/g) and productivity (0.19 g/Lh) were verified after 72 hours.

A cross-sectional study on HIV/AIDS was carried out in 108 outpatients from the university hospital of the Federal University of Mato Grosso do Sul, Campo Grande, Brazil, from July to December 2008, to investigate latent tuberculosis infection using the tuberculin skin test (TST). The prevalence of positive results was 13.9 percent. The CD4+ T cell count (p = 0.091) and the diagnosis time (p = 0.010) were statistically significant when compared with TST positivity. In the cohort of HIV/AIDS patients who had latent tuberculosis infection, the median diagnosis time was eight years. Undetectable viral load presented significant association (p = 0.046) with tuberculosis infection. The fact that numerous individuals with HIV/AIDS infection presented a negative reaction to the tuberculin skin test is probably related to alterations in the cellular immune response induced by HIV infection. The tuberculin test is a useful tool for the detection of latent tuberculosis infection and should be performed in all HIV/AIDS individuals at the time of the diagnosis and on a yearly basis, if negative. Both the early identification of the tuberculosis infection by the tuberculin skin test at the moment of immunological restoration and chemoprophylaxis in infected individuals are mechanisms to control HIV/AIDS and tuberculosis coinfection.

ABSTRACT

We developed a database system for collaborative HIV analysis (DBCollHIV) in Brazil. The main purpose of our DBCollHIV project was to develop an HIV-integrated database system with analytical bioinformatics tools that would support the needs of Brazilian research groups for data storage and sequence analysis. Whenever authorized by the principal investigator, this system also allows the integration of data from different studies and/or the release of the data to the general public. The development of a database that combines sequences associated with clinical/epidemiological data is difficult without the active support of interdisciplinary investigators. A functional database that securely stores data and helps the investigator to manipulate their sequences before publication would be an attractive tool for investigators depositing their data and collaborating with other groups. DBCollHIV allows investigators to manipulate their own datasets, as well as integrating molecular and clinical HIV data, in an innovative fashion.

ABSTRACT

A imunização de hospedeiros vertebrados com componentes derivados de vetores pode se constituir numa estratégia alternativa para o controle de doenças transmitidas por insetos. No presente estudo avaliamos o efeito de anticorpos antiflebótomos sobre alguns parâmetros biológicos de fêmeas de Lutzomyia longipalpis, vetor de leishmaniose visceral. Coelhos foram imunizados com extratos de tubos digestivos de fêmeas alimentadas com açúcar (GS), fêmeas alimentadas com sangue (GB), carcaças de fêmeas alimentadas com açúcar (CS) ou carcaças de fêmeas alimentadas com sangue (CB), e coelho imunizado por repetidas picadas de fêmeas de flebótomos (BITE). Os soros imunes de coelhos apresentaram títulos aumentados quando comparados com os soros pré-imunes, e bandas específicas foram detectadas por meio de Western Blot. A análise dos parâmetros biológicos revelou um decréscimo na fecundidade no grupo de fêmeas alimentadas em coelho imunizado com GB e BITE. A longevidade e a mortalidade foram estudadas em fêmeas com postura (paridas) e fêmeas sem postura (nulíparas). Fêmeas nulíparas que se alimentaram em coelho imunizado por repetidas picadas morreram em maior percentual. A análise da mortalidade, após a postura dos ovos, revelou um pico no quinto dia em todos os grupos, mas em fêmeas que se alimentaram em coelho submetido a repetidas picadas, foi antecipada para o terceiro dia.

Thalidomide is a selective inhibitor of tumor necrosis factor-alpha (TNF-alpha), a cytokine involved in mycobacterial death mechanisms. We investigated the role of this drug in the functional activity of alveolar macrophages in the presence of infection induced by intranasal inoculation of Mycobacterium avium in thalidomide-treated and untreated adult Swiss mice. Sixty animals were inoculated with 5 x 10(6) M. avium by the respiratory route. Thirty animals received daily thalidomide (30 mg/kg mouse) and 30 received water by gavage up to sacrifice. Ten non-inoculated mice were used as a control group. Lots of animals from each group were evaluated until 6 weeks after inoculation. Infection resulted in an increased total number of inflammatory cells as well as increased activity of pulmonary macrophages. Histologically, intranasal inoculation of bacilli resulted in small mononuclear infiltrates located at the periphery of the organ. Culture of lung fragments revealed the presence of bacilli only at the beginning and at the end of the experimental period. Thalidomide administration did not affect the microbiological or histological features of the infection. Thalidomide-treated and untreated animals showed the same amount of M. avium colonies 3 weeks after infection. Although it did not affect bacillary clearance, thalidomide administration resulted in a decreased percent of spread cells and release of hydrogen peroxide, suggesting that factors other than TNF-alpha play a role in the killing of mycobacteria by alveolar macrophages. Thalidomide administration also reduced the number of spread cells among resident macrophages, suggesting a direct effect of the drug on this phenomenon.

ABSTRACT

Se analizó el impacto de las enfermedades fúngicas en las pérdidas de postcosecha de papaya en la fase de comercialización, evaluando la influencia de la temperatura en la manifestación de las enfermedades de postcosecha y del binomio temperatura-humedad relativa en la severidad de la pudrición causada por Lasiodiplodia theobromae. Cuatro supermercados, en la cuidad de Recife (Estado de Pernambuco, Brasil), fueron analizados en cuanto al impacto de las enfermedades fúngicas postcosecha en las pérdidas de frutos de papaya. Estas fueron las principales causas de descarte, correspondiendo un 98 por ciento al Cultivar Formosa y 90,5 por ciento al Cultivar Sunrise Solo. Los principales hongos involucrados en el proceso de infección fueron Colletotrichum gloeosporiodes, L. Theobromae y Rhizopus stolonifer, agentes causales de la antracnosis, pudrición peduncular y pudrición acuosa, respectivamente. Al analizar la influencia de la temperatura (5, 15 y 30°C) en los frutos con aspecto sano, se constató que las principales enfermedades fueron las mismas observadas en los frutos descartados. A temperaturas de 5 y 15°C, la antracnosis presentó una mayor incidencia en relación con las otras enfermedades, mientras que la pudrición peduncular fue constatada solamente a 30°C, con incidencia superior a las demás. En el análisis de la influencia de la temperatura y la humedad sobre la severidad de la pudrición causada por L. theobromae, no se observaron síntomas de la enfermedad en frutos en humedad relativa inferior o igual al 90 por ciento. En presencia de elevada humedad, sólo a partir de 25°C fue constada la incidencia de la enfermedad, con un aumento en la severidad de la misma con la elevación de la temperatura y del período de humedad relativa.

ABSTRACT

Molecular studies of insect disease vectors are of paramount importance for understanding parasite-vector relationship. Advances in this area have led to important findings regarding changes in vectors' physiology upon blood feeding and parasite infection. Mechanisms for interfering with the vectorial capacity of insects responsible for the transmission of diseases such as malaria, Chagas disease and dengue fever are being devised with the ultimate goal of developing transgenic insects. A primary necessity for this goal is information on gene expression and control in the target insect. Our group is investigating molecular aspects of the interaction between Leishmania parasites and Lutzomyia sand flies. As an initial step in our studies we have used random sequencing of cDNA clones from two expression libraries made from head/thorax and abdomen of sugar fed L. longipalpis for the identification of expressed sequence tags (EST). We applied differential display reverse transcriptase-PCR and randomly amplified polymorphic DNA-PCR to characterize differentially expressed mRNA from sugar and blood fed insects, and, in one case, from a L. (V.) braziliensis-infected L. longipalpis. We identified 37 cDNAs that have shown homology to known sequences from GeneBank. Of these, 32 cDNAs code for constitutive proteins such as zinc finger protein, glutamine synthetase, G binding protein, ubiquitin conjugating enzyme. Three are putative differentially expressed cDNAs from blood fed and Leishmania-infected midgut, a chitinase, a V-ATPase and a MAP kinase. Finally, two sequences are homologous to Drosophila melanogaster gene products recently discovered through the Drosophila genome initiative

ABSTRACT

En la central de abastecimiento y en tres ferias libres de la ciudad de Recife, PE(Brasil), se analizaron cada tres meses, durante un año, frutos de aguacate (palta), limón, maracuyá-amarillo, pimentón y tomate para detectar la presencia de enfermedades fúngicas. En cada muestra fueron evaluadas 20 unidades de cada tipo de fruto, en cinco puntos diferentes por sitio de colecta, totalizándo 100 frutos/punto de comercialización, la incidencia de enfermedades fúngicas en los frutos de aguacate fue de 38,52 porciento, en maracuyá de 15,48 porciento, en limón de 17,55 porciento, en pimentón de 1,58 porciento y en tomate de 6,5 porciento. El hongo detectado con mayor frecuencia en los frutos de aguacate fue sphaceloma perseae (20,0 porciento), en limón sphaceloma fawcetti (6,2 porciento), en maracuja cladosporium herbarum (10,3 porciento) en pimentón alternaria alternata (0,7 porciento) y en tomate geotrichum candidum (3,2 porciento). Los altos valores de incidencia de enfermedades fúngicas detectados, indican la necesidad de adoptar procedimientos antes y después de la cosecha, para reducir los riesgos de infección, lo que promovería la reducción significativa de pérdidas de postcosecha

Tetralogy of Fallot was diagnosed in a 30 month-old Cocker Spaniel female, through physical, radiographic, electrocardiographic, and echocardiographic exams. The animal was submitted to a palliative surgical correction, the Blalock-Taussig technique, and 46 months after the surgery, no signs of cardiopulmonary decompensation were observed

O objetivo deste trabalho foi avaliar a longevidade dos espermatozóides provenientes da fraçäo rica e da fraçäo completa de ejaculados de jumentos, preservados em dois diluidores. As duas amostras seminais foram diluídas nos dois diluidores (Kenney e Baken modificado) e preservadas a 5§C. Sêmens de cinco jumentos foram coletados com auxílio de vagina artificial de fundo aberto, uma vez por semana e por cinco semanas consecutivas. Os dois primeiros jatos do ejaculado (fraçäo rica) foram coletados separadamente do restante do ejaculado. Amostras de ejaculado completo foram obtidas misturando proporcionalmente parte da fraçäo rica e parte do restante do ejaculado. Amostras de sêmen foram, em seguida, misturadas 1:1 com cada diluidor e mantidas em temperatura ambiente até a avaliaçäo da concentraçäo. A diluiçäo final teve por objetivo a obtençäo de 50 x l0 elevada a sexta potência espermatóides por ml. O sêmen diluído foi resfriado até 5§C numa taxa de -0.6§C/min. A motilidade total e progressiva e o vigor foram avaliados no final da diluiçäo, após resfriamentos e a cada 24 horas até a queda da motilidade total abaixo de l0 por cento. A morfologia espermática foi avaliada por microscopia de contraste de fase em amostras coletadas após a diluiçäo, e nos dias 3, 6 e 9 pós-coleta. As médias foram comparadas pelo teste t de Student. Näo houve diferença na longevidade dos espermatozóides entre a fraçäo seminal rica e a fraçäo completa com ambos os diluidores testados. Observou-se que os espermatozóides, tanto da fraçäo rica como da completa, foram melhor preservados no meio Baken modificado (base de gema de ovo) do que no meio Kenney (base de leite desnatado)

ABSTRACT

The aminopeptidase activity of a homogenate of rabbit kidney treated with Triton X-100 was measured using L-aminoacyl-2-naphthylatmides (AA-NA). After gradient elution ion-exchange chromatography, four peaks of aminopeptidase activity were eluted. The enzyme eluted at 450 muS containing 33.5 per cent of the activity towards Arg-NA was applied to a Superdex 75 column and presented only one protein band on 10 per cent SDS-polyacrylamide gel electrophoresis. This enzyme has an apparent molecular mass of 78 kDa, is five-fold activated by 0.15 M NaCl and the highest Vmax/Km ratio was obtained with Arg-NA. Enzyme activity was inhibited 100 per cent by 0.13 mM sodium p-hydroxymercuribenzoate, 20 per cent by 0.75 mM EDTA and 100 per cent by 0.66 mM ophenanthroline. Puromycin and bestatin behaved like competitive inhibitors with a Ki of 0.60 mM and 5.0 muM, respectively.

ABSTRACT

Estudaram-se no presente trabalho 28 casos de cardiomiopatia (CM) do Cocker Spaniel, com a finalidade de determinar as características epidemiológicas, clínicas e etiológicas da doença. Para tanto, os animais foram avaliados por meio de anamnese, exame físico, exame radiológico e eletrocardiograma. Dos 28 animais, 13 eram machos e 15 fêmeas, com idade variando entre 18 e 144 meses. As manifestaçöes clínicas mais freqüentemente associadas com este tipo de CM foram intolerância ao exercício, tosse, dispnéia e engasgo, observados, respectivamente, em 23, 14, 7 e 7 casos. O exame radiográfico realizado em 12 animais revelou como alteraçöes mais comuns: desvio dorsal de traquéia (8/12), aumento da parede cranial e direita do coraçäo (7/12), cardiomegalia global (5/12) e aumento do interstício pulmonar (4/12). O exame eletrocardiográfico revelou freqüência cardíaca variando entre 60 e 180 b.p.m. Os ritmos cardíacos mais freqüentes foram arritmia sinusial associada a marcapasso migratório (11/28), ritmo sinusal normal (6/28) e arritmia sinusal (5/28); o eixo cardíaco no plano frontal mostrou-se dentro da normalidade na maioria dos casos (27/28); o eletrocardiograma revelou, ainda, sobrecarga ventricular direita (28/28) e sobrecarga ventricular esquerda (20/28)

ABSTRACT

A evidencia da transmissao extraflorestal da leishmaniose cutaneo-mucosa na regiao do Vale do Ribeira ensejou o presente estudo epidemiologico prospectivo, visando avaliar a atividade enzootica de L. (V.) braziliensis. A pesquisa paratisologica da infeccao natural em pequenos mamiferos e populacao canina foi complementada com o teste de imunofluorescencia indireta (IFI) para caes e captura de flebotomineos em ambiente florestal e peridomiciliar. A positividade para o teste sorologico e exame parasitologico somente foi observada para caes residentes e com taxas de 5,6 e 2,4 por cento, respectivamente. Entre animais silvestres e sinantropicos capturados, destacam-se os pertencentes a Oryzomys (Oligoryzomys) e Rattus rattus, ambos assinalados em proporcoes equivalentes (29,3 por cento) em ambiente peridomiciliar. Foram capturados apenas 166 exemplares femininos de Lutzomyia intermedia, fato atribuido a borrifacao das habitacoes humanas e anexos com DDT. No contexto epidemiologico mais amplo, discute-se a fragilidade do ciclo extraflorestal da L. (V.) braziliensis; o papel do cao e de pequenos mamiferos, como fonte de infeccao domiciliar, alem de analisar o potencial deles na dispersao do parasita na area estudada

ABSTRACT

The maximal expiratory flow-volume curve was used to evaluate existing physiological abnormalities in forty patientes with chronic Chagas'Disease. All patients had megaoesophagus with or without megacolon, some had ECG evidence of cardiomyopathy but none had clinic signs of cardiac failure. The 40 patients were studied in four groups: I)The total group with or without cardiomyopathy. II) 24 patients with associated cardiomyopathy. III) 16 patients without cardiomyopathy. IV) 17 non smoking patients with or without cardiomyopathy. The results indicated that there was: a) a dmininution of the forced vital capacity (FVC) in group I, II and IV; b) a diminution of maximal expiratory flow (Vmax) in group I and II; c) a diminution of the maximal expiratory flow rates at 50% and 25% of the expired vital capacity (Vmax50 and Vmax25) in all four groups. The alterations of the first-second forced expired volume (FEV1) were not significant in any group. Diminution of FVC and Vmax seen to be dependent upon the presence of associated cardiomyopathy while diminution of the Vmax50 and Vmax25 was a constant finding, independent of the presence of cardiopathy
